what is the difference between error and defect?

error is the mistake in a program, but the missed some functional point or feature are called the defects.an error can make the big defects in future.

manual testing can replace by automation?

no, manual testing not replaced by automation testing. because if the person not knowing write the test case , how he/she execute the test script.
